Treatment of corneal epithelial wounds in dogs using basic fibroblast growth factor

چکیده
An experimental study examined the effect of basic fibroblast growth factor (bFGF) on the healing of corneal epithelial wounds in dogs. A corneal wound was made on one eye from each of 40 dogs with a corneal trephine (6 mm diameter). Four concentrations of bFGF (0, 0.1, 0.5, and 1.0 μg/ml) were applied to the affected eyes three times daily. Fluorescein staining was used to assess the closure of the corneal epithelial wounds. The morphological characteristics were determined on histological examination. The wound healing rate was significantly greater in the bFGF-treated group compared with controls 1, 3, 5, and 7 days (P < 0.01) after the topical administration of bFGF. Both 0.5 and 1.0 μg/ml bFGF increased the wound healing rate significantly (at Days 3 and 5, P < 0.05) compared to 0.1 μg/ml bFGF. Moreover, two control cases still showed poor healing 10 days after the corneal wound. None of the eyes developed corneal clouding or neovascularization during the experiment. The histological examination showed more epithelial layers, a more regular rearrangement, and fewer inflammatory cells in the epithelium of the bFGF-treated group; the epithelium was reconstructed more quickly in the bFGF-treated group compared with the control group. These results suggest that bFGF promotes canine corneal epithelial wound healing effectively, making bFGF suitable for curing canine corneal epithelial wounds.

The purpose of this study was to evaluate the effects of different time combinations of dexamethasoneand acetylcysteine on experimentally induced corneal ulcers in dogs. Experimental corneal wounds werecreated surgically to the anterior one third of the cornea in the center of all eyes of 15 mixed breed dogs. The eyes were divided into five groups according to planned post-operative medications...
متن کاملCorneal neovascularization after excimer keratectomy wounds in matrilysin-deficient mice.
PURPOSE Matrilysin, matrix metalloproteinase (MMP)-7, is upregulated in the corneal epithelium during wound healing after excimer keratectomy wounds. The purpose of this study was to determine the role of matrilysin in maintaining corneal avascularity during wound healing. METHODS Matrilysin-deficient mice (n = 17) and their age-matched wild-type littermates (n = 18) were treated with 193 nm ...
متن کاملEGF, EGF receptor, basic FGF, TGF beta-1, and IL-1 alpha mRNA in human corneal epithelial cells and stromal fibroblasts.
We sought to determine whether human corneal epithelial cells and stromal fibroblasts synthesize messenger RNAs (mRNA) coding for epidermal growth factor (EGF), EGF receptor, basic fibroblast growth factor (basic FGF), transforming growth factor-beta 1 (TGF-beta 1), and interleukin-1 alpha (IL-1 alpha). Total cellular RNA was extracted from cultured stromal fibroblasts and ex vivo and cultured ...

The polymerase chain reaction (PCR) was used to demonstrate that rabbit corneal epithelial cells produce messenger RNAs coding for epidermal growth factor (EGF), basic fibroblast growth factor (FGFb), and transforming growth factor beta-1 (TGF beta 1) ex vivo and in primary culture. EGF, FGFb, and TGF beta 1 mRNAs were detected in central and peripheral ex vivo epithelial tissue in wounded and ...
